  • 摘要:流行病学研究表明,空气细颗粒污染物(PM2.5)的暴露与过敏性疾病有一定的联系;然而,PM2.5暴露与过敏性疾病之间的关系尚未完全阐明,特别是室内环境中PM2.5涉及到过敏或非过敏的作用不详.为了比较研究过敏与非过敏儿童室内PM2.5的细胞毒性,在武汉市洪山区10户家庭室内进行了为期3个月的采样,分别收集过敏与非过敏儿童的室内PM2.5.采用有机/元素碳测定仪对二者PM2.5成分中的含碳组分进行了分析,并通过检测昆明小鼠巨噬细胞的形态及吞噬功能影响、细胞活力、乳酸脱氢酶(LDH)漏出率等指标,来检测PM2.5暴露所致的细胞毒性.结果表明,高剂量(200 μg·mL-1)PM2.5暴露对小鼠巨噬细胞的形态及吞噬功能会产生不利的影响;与非过敏儿童的室内PM2.5暴露组相比,过敏儿童的室内PM2.5暴露组诱导巨噬细胞产生的毒性作用更明显.细胞体外测试结果提示:在相同PM2.5暴露剂量下,引起儿童过敏症的室内PM2.5成分具有重要影响.
  • Abstract:Epidemiological studies have shown exposure to air fine particulate matter (PM2.5) is associated with allergic diseases. However, the association between PM2.5 exposure and allergic diseases has not yet been fully elucidated, particularly the role of PM2.5 from indoor environment involved in allergy or non-allergy is unknown. To compare cytotoxicity of indoors PM2.5 samples, which collected from allergic or from non-allergic children's homes, we conducted a 3-month sampling in 10 households in Hongshan District, Wuhan City Center. The carbon composition of indoor PM2.5 was analyzed by an OC/EC analyzer (Sunset Laboratory Inc.), and the cytotoxic indicators included the phagocytic function, cell viability and lactate dehydrogenase (LDH) leakage rate of KM mice peritoneal macrophage exposed by the indoor PM2.5. Experimental results showed that ①After exposure to PM2.5 (200 μg·mL-1), the shapes and phagocytic function of the macrophages are significant change; ②The indoor PM2.5 exposure group from the homes of children with allergic symptoms induced macrophages produce more serious toxic effects than that of healthy ones. The results of this vitro study suggest that the children's allergic symptoms are potentially associated with the indoor PM2.5 compositions and their contents.
